Can back up be done through Mobile Data?

No -- as in my previous reply "It requires wi-fi."
You may want to give feedback to Apple about giving the mobile data option: Feedback - iCloud - Apple
I am aware of the Wi-Fi but very interested to know if perhaps mobile data can be used when Wi-Fi isn't available.
